Step into the world of polarized light microscopy

Polarizing microscope is a microscope used to study so-called transparent and opaque anisotropic materials.  Where there is birefringence of the material, under the polarizing microscope can be clearly distinguished, of course, these substances can also be observed by staining, but some are impossible, and must use the polarizing microscope.

Microscopic examination of a substance by changing ordinary light to polarized light to identify whether it is monorefringent (isotropic) or birefringent (anisotropic).  Birefringence is the basic property of crystals.  Therefore, polarized light microscope is widely used in the fields of minerals, chemistry, biology and botany.

2. Basic principles of polarized light microscopy  

The principle of polarizing microscope is more complex, which will not be introduced here. Polarizing microscope must have the following accessories: polarizing mirror, polarizing mirror, compensator or phase piece, special stress-free objective lens, rotating loading platform.  


3. Methods of polarizing microscopy  

Normal microscopy: also known as aberration-free microscopy, which is characterized by the use of low-power objective lens, no Bertrand lens, the subject can be directly studied with polarized light.  At the same time, in order to make the lighting aperture smaller, push the condenser lens.  Orthophotoscopy is used to check the birefringence of objects.  


Conical microscopy: Also known as interferometry, the study of interference patterns produced when polarized light interferes. this method is used to observe uniaxial or biaxial properties of objects.  In this method, a strongly convergent polarized beam is used for illumination.  


4. Device requirements for polarizing microscope 

Light source: it is best to use monochromatic light because the speed, refractive index, and interference of light vary with wavelength.  General microscopic examination can use ordinary light.  


Eyepiece: Eyepiece with crosshair.  


Converging lens: To achieve parallel polarization, a roll-out converging lens should be used.

Bertrand lens: An auxiliary lens in the path of a condenser, which amplifies all the primary phases of an object into secondary phases.  It enables the eyepiece to view the flat pattern formed in the focal plane behind the objective lens.  


5. Requirements for polarizing microscopy  

The center of the platform is coaxial with the optical axis.  

The polarizing mirror and the polarizing mirror should be in an orthogonal position.  

The film should not be too thin.  


6. What should be paid attention to in the use of polarized light microscope?  

First, after turning on the polarizing microscope power supply, if it is not used temporarily, you can turn the microscope light to dark. Before turning off the polarizing microscope power supply, turn the microscope light to dark every time. After the light box cools completely (about 15 minutes later), the power supply wire will be rolled up, without frequently switching the polarizing microscope power supply.  


Second, intraoperative adjustment was performed under aseptic conditions.  


Third, take good care of the power cord and the foot control switch so as not to damage the polarizing microscope related devices, and return to the position in time after use.  


Fourth, pay attention to keep the optical lens clean, fingers should not touch the optical surface, each such as the lens surface dust can be clean brush or brush to play.  If there is oil pollution, you can use absorbent cotton with a small amount of ethanol mixture (30% ethanol) wipe, wipe do not let the mixture into the lens, so as not to deglue the lens.

Fifth, when polarized light microscope is used, optical fiber is not easy to be subjected to large bending force. When it needs to be bent, the bending radian is small, so as not to break.  


Sixth, polarized light microscope has two groups of light source lamp 1 and lamp 2, can be switched at any time when using, the bulb switch rotary table must be switched in place, otherwise it will affect the illumination degree.  The bulb should be changed after 30 minutes of shutdown to avoid high temperature burns.  


Seventh, when using and moving the polarizing microscope, hold the handle of the frame and push it slowly. Do not place the bearing arm, spring arm and the head on the back side of the polarizing microscope panel at the same time to avoid danger caused by tipping.  


Eighth, the use and storage of polarized light microscope place should be kept clean, pay attention to dust, moisture, acid and alkali corrosion and other corrosive substances.  When polarizing microscope is used or not used for a long time, dust cover should be covered.
